Obesityisamajorcancerriskfactorwhichinducessystemicperturbationsleadingtodyslipidemia,insulinresistance,alteredhormonelevels,andinflammation.However,ithasnotyetbeenreportedhowchangesinsystemicmetabolisminducedbyobesityaffectimmunecellsinthelocaltumormicroenvironment.Thus,inthispaper,theauthorsinvestigatedthatHFDcanacceleratetumorgrowth.FurtheranalysisshowedthatHFDespeciallyaffectedCD8+Tcellnumbersandfunctioninthetumor.Usingsinglecellsequenceanalysis,theyfoundthatlymphocytesweresignificantlyreducedinanimalsonHFDandnaiveorunstimulatedTcellstendedtobeenrichedwithHFDdisplayedalteredexpressionofmajorcentralcarbonmetabolicpathways.Usingmetabolic,proteomic,andgenomicapproaches,aswellasmultiplexedtissueimaging,theirdatarevealedMetabolicRemodelinginTumorswithHFDthatenhancedfattyaciduptakebytumorcellsleaveTcellsdeprivedoffattyacidsintheTME.Inaddition,theydemonstratedthatpreventingHFD-inducedmetabolicrewiringbymodulatingPHD3expressionintumorcellscouldcontrolfattyacidavailabilityintheTME,finallyaffectingCD8+TcellresponsesandtumorgrowthonHFD.Insummary,thispaperoffersapossibilitythatimplicationsfortherapeuticinterventionstargetingcancermetabolismmayhaveimpactsontheantitumor-immunityandprecisionmedicine.
